NCT07885267

Brief Summary

This is a first-in-human, open-label, multicenter Phase 1 study of AT-0174, an investigational oral dual TDO2/IDO1 inhibitor. It is being conducted in adults with advanced solid tumors that have progressed on, or are not suitable for, standard therapy. The main goals are to evaluate the safety and tolerability of AT-0174 and to identify a recommended dose for further study.

Outcome Measures

Primary Outcomes (3)

  • Number of participants with dose-limiting toxicities (DLTs)

DLTs graded per NCI CTCAE v5.0.

    Cycle 1 (28 days)

  • Number of participants with treatment-emergent adverse events

Adverse events graded per NCI CTCAE v5.0.

From first dose through 30 days after last dose

  • Recommended Phase 2 dose (RP2D) of AT-0174

Determined from safety and tolerability data.

Through study completion, approximately 33 months

Secondary Outcomes (1)

  • Percent change from baseline in plasma kynurenine (KYN) and tryptophan (TRP) concentrations

    Baseline (pre-dose) through Cycle 4 (28-day cycles)

Study Arms (1)

AT-0174

EXPERIMENTAL

Participants receive AT-0174 orally once daily in 28-day cycles at assigned dose levels.

Drug: AT-0174

Interventions
